This workshop will introduce the basic methodology of handling arbuscular myorrhiza fungi, their axenic culture and approaches to their transient transformation. Topics will include: (1) In vitro AM cultivation; Isolation of spores, hyphae and nucleic acids from AM fungi. (2) Transformation protocols for transient expression of reporter genes in AM fungi, suitable vectors for A. tumefaciens and bombardment-based transformation, selection markers. (3) Micro-dissection methods to isolate fungal structures from within the root. This course is in high demand and will also be open to participants outside this SPP.
